Surmodics IVD offers several TMB stop solutions, including our most recently developed TMB stop solution, BioFX™ 450 nm Liquid Nova-Stop Solution for TMB Microwell Substrates (Product Code: NSTP). This TMB stop solution offers assay developers maximum performance, and minimal signal drift compared to sulfuric or hydrochloric acid. Absorbance is read in the yellow spectral range (450 nm) and can be read up to 3 hours after the TMB reaction has been stopped using the Nova-Stop Solution. In addition, this TMB stop solution offers assay developers a safer alternative as it is non-corrosive to both skin and eyes.

Intended Use of Stop Solution for TMB Substrate

In addition to our TMB stop solutions, Surmodics IVD provides assay developers with a range of best-in-class TMB substrates. We offer a wide variety of TMB substrates at varying kinetic rates, including TMB conductivity, super sensitive, one component, extended range, double slow, among others. These ready-to-use TMB substrates offer up to 4 years of shelf life stability and prolonged room temperature stability and increased signal-to-noise ratios and ability to quantitate through the desired assay range. In addition, they provide negligible backgrounds yielding lower error through any assay system.

In observing how the TMB substrate and stop solution interact, peroxidase produces a soluble blue reaction when combined with the TMB Substrate, adding a TMB stop solution stabilizes the color development and readings.

TMB stop solutions terminate the reaction of the substrate. In this case, with the use of the TMB stop solution, the TMB is optimal for ELISA applications. The solution allows the TMB products to attain the expected color intensity, which indicates a complete analyte reaction.

The immobilized horseradish peroxidase reacts with the TMB products to form a blue solution that turns yellow upon adding the TMB stop solution. The reaction becomes stable at an absorbance maximum of 450 nm, and this extends the measurement period.

Applications of Stop Solution for TMB Substrate

The TMB stop solution is critical in application since it allows the substrate to undergo a complete reaction. The use of the solution on the enzyme-substrate in simple terms is for bringing about a change of color. The shift from blue to yellow is vital for the reactants to form the ultimate product for the process to stop. The yellow indicates that there is no further reaction taking place, hence the formation of the final product. The stop solution also changes the pH of the substrate to bring out the color change and make it optimal for ELISA’s.
