Prolonged shelf life. BioFX™ One Component HRP Microwell Substrate (TMBW) maintains 96% activity for over 4 years.
Thirty-two lots spanning 55 months of production were collected and analyzed in a commercial ELISA assay. The color development (Abs @650) was monitored for 30 minutes to determine the kinetic rates across 2 different plates (3 repetitions per substrate on each plate) and the results were normalized to a new lot of TMBW (Age of lot=0 months).

An IL-6 immunoassay (R&D Systems) was run with TMBHD, TMBW, and other high sensitivity TMB substrates to compare limit of detection and sensitivity. A standard curve was prepared from 100fg/ well to 6.25 fg/well of IL-6. Signal is plotted as the 450nm OD of each well minus the background (zero standard) value. Detection limit was determined by Student’s T test (p < 0.05) at the lowest possible tested analyte concentration.

An HRP standard was added to a rabbit anti-HRP antibody coated microplate, incubated and washed. 7 different lots of TMBW were added to designated microwells and absorbance was read every 5 minutes of the kinetic assay.
BOOSTING SENSITIVITY
Surmodics TMB Enhanced HRP Membrane Substrate (ESPM) provides extended dynamic range with reduction of wash out at higher peroxidase concentrations. ESPM demostrates similar sensitivity to most mid-to-high level chemiluminescent subtrates without the costly equipment needed for detection.
Dot blots were prepared with a 1:3 serial dilution of an HRP conjugated antibody starting at 1,000 pg/well, conjugates were incubated and then washed. Membranes were placed in substrate, incubated and read.
